چکیده

At the synapse the specialized preand postsynaptic membranes are connected via a structure called the synaptic junction. In order to understand the nature of the synaptic junction or connection, it is necessary to determine which molecules are associated with the surfaces of the specialized preand postsynaptic membranes and which are present in the cleft itself. Cytochemical evidence indicates the possible presence of carbohydrates in the cleft. The cleft area stains with ruthenium red and periodic acid-Schiff stain (Rambourg and Leblond, 1969; Bondareff, 1967). Recently Matus et al. (1973) suggested that concanavalin A (Con A) binding sites may be present in the synaptic cleft. Since Con A binds to mannopyranosyl or glucopyranosyl receptors, the cleft would appear to contain these sugars or closely related molecules. At present it is not known whether Con A sites are soluble or membrane bound and nothing is known about the type of molecule to which the lectin binds. In this paper we describe the organization of Con A binding sites in the synaptic cleft of isolated synaptosomes and synaptic junctional complexes (SJC) by the use of ferritin-Con A conjugates and quantitate .the binding and nature of the receptors by the use of radioactive Con A. We confirm the observations of Matus et al. (1973) and demonstrate that within the cleft a portion of the Con A binding sites are present on the external surface of the postsynaptic membrane overlying the postsynaptic density (PSD). These sites appear to be mainly glycoproteins. In contrast to the surface membrane the underlying PSD binds very little, if any, Con A conjugate.

Characterization of spontaneous network-driven synaptic activity in rat hippocampal slice cultures

A particular characteristic of the neonatal hippocampus is the presence of spontaneous network-driven oscillatory events, the so-called giant depolarizing potentials (GDPs). GDPs depend on the interplay between GABA and glutamate. Early in development, GABA, acting on GABAA receptors, depolarizes neuronal membranes via a Cl- efflux. Glutamate, via AMPA receptors, generates a positive feedback n...

The Role of Adrenergic Receptors on Neural Excitability and Synaptic Plasticity: A Narrative Review

Adrenergic receptors have an important role in neural excitability and synaptic plasticity. Despite a lot of studies on these receptors, their exact role in brain disorders accompanied with hyperexcitability has not been determined. There are also controversies on their role in synaptic plasticity. In this review article, the important studies done in this regard have been reviewed to achieve a...

P24: The Role of Ionotropic Glutamate Receptors in the Induction of LTP

Long-term potentiation (LTP) is a reflection of synaptic plasticity that has an important role in learning and memory. LTP is a long-lasting increase of synaptic activity due to enhancement of excitatory synaptic transmission after a high-frequency train of electrical stimulation. The role of α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors in excitatory synaptic tran...

A novel synaptic junction preparation for the identification and characterization of cleft proteins

Identification of synaptic cleft components has been hampered by the lack of a suitable preparation enriched in synaptic junctions devoid of adjoining peripheral membranes. Prior strategies for the isolation of synaptic junctions, relying on detergents for the removal of peripheral membranes, resulted in substantial loss of membranes lining the cleft. Here, a novel, detergent-free method is des...
